The total aneuploidy rate for our sample set was calculated to be 20.8%, closer to the estimate of 20-25% from other sources. We are carrying out a study to determine the mechanisms leading to aneuploidy, and therefore implantation/pregnancy failure, in this cohort of patients. Furthermore, such events are thought to occur with equal frequency at any maternal age, in contrast to aneuploidy, which is associated with advanced maternal age. These aneuploidy data may also have been influenced by differences in the bioavailability of tamoxifen among species and in target organ specificity of tamoxifen activity. The observed aneuploidy (hypo- or hyperploidy) can be due either to defective chromosome segregation or simply to technical errors (loss of chromosomes, erroneous counting). No comparative data were available regarding the incidence of aneuploidy. It seems reasonable to suggest that pyelectasis increases the prior risk of aneuploidy to some degree. In this model, investigators characterized different outgrowth lines with and without aneuploidy. First trimester screening for fetal aneuploidy: biochemistry and nuchal translucency. Disruption of the spindle leads to abnormal chromosomal segregation predisposing to aneuploidy and hence apoptosis at the pre-implantation embryonic stage. Evidence of several mechanisms leading to aneuploidy was provided. The median cleft, occurring in only 1% of facial clefts overall is nearly always associated with additional structural anomalies and aneuploidy (particularly trisomy 13). Detection rates for aneuploidy would be expected to be higher therefore than in the general population. The most common abnormalities in these studies were aneuploidy and polyploidy. From these studies, it is suggested that screening earlier in pregnancy will detect other pregnancy-related abnormalities, in addition to aneuploidy. Meiotic spindle is crucial for the configuration of chromosomes, hence disruption of spindles causes abnormal chromosome distribution, which may cause aneuploidy after fertilization. In contrast, only 17% of oocytes from the younger age group exhibited aneuploidy. In each cell analysed, the number of chromosomes and/or chromatids was counted at x1250 magnification to provide data for calculating aneuploidy. |
